Ultrastructurally Pathological Study of Renal Tubular Apoptosis Following Unilateral Ureteral Obstruction

Objectives

Acute unilateral urethral obstruction may likely make some changes in expression inducer genes of apoptosis and engaged growth factors, e.g., EGF, IGF-1 and b FGF, and cause induction of apoptosis in renal mesangial and proximal tubular cell. We made unilateral urethral obstruction (uuo) in canine (animal) model, thereby the apoptosis changes induced by uuo experienced in canine model and evaluated by ultrastructural pathology.

Material & Methods

Totally ten dogs, 5 of them were surgically performed complete uuo, and the rest were as control group. Seven days later, renal samples were biopsies from both (treatment and control) groups and associated section for light and Electronic Microscopes (LM&EM) were prepared and assessed.

Results

LM studies revealed for treatment group: formation of round and oval cytoplasmic masses with or without nuclear basophilic bodies with chromatin crescents in renal tubular cells. EM studies also showed nuclear chromatin formations as osmophilic granules with high dense nucleus and fragmentation in these cells of treatment group. In contrary, in the control group, there were no significant changes of apoptosis, due to uuo.

Conclusion

This study demonstrated that acute uuo in canine (animal) model can induce cell apoptosis by making some changes in growth factors, which affect on expression of genes producing growth factors such as EGF. And also in this case the most important of them is expression of sulfate glycoprotein-2 (SGP-2), which as an important genetic marker can definitely affect the induction of apoptosis during uuo. The abovementioned factors influence were shown in our study as ultra structural pathologic morphological manifestations.
